چکیده

With diabetes, skeletal muscle mitochondrial quality control (mitochondrial fusion, fission & macro-autophagy) is impaired. Geranylgeraniol (GG) shown to have a protective effect on preventing damage and health; however, the of GG diabetic model not known. PURPOSE: To determine cross-sectional area (CSA) in rats. METHODS: Thirty-five Sprague-Dawley rats were divided into three diet groups: (CON), high-fat with 35 mg/kg body weight streptozotocin (HFD), HFD 800 (GG). Due limited sample, total 21 (CON: n = 7; HFD: GG: 7) rats’ samples used for this report. The soleus muscles harvested after 7-weeks feeding analyzed OPA1, MFN2, DRP1, pDRP, PINK1, Parkin, LC3A, LC3B protein content using western blot analysis. Muscle CSAs assessed Image J. RESULTS: A significant (p < 0.05) condition was observed contents CSA. For (0.21 ± 0.08) had lower MFN2 than CON (0.43 0.04; p 0.007) (0.65 0.08; 0.010). fission, (0.26 0.07) DRP1 (0.59 0.07; 0.019). macro-autophagy, (1.08 0.28) LC3A (2.81 0.55; 0.028) (3.99 0.57; 0.010); whereas (0.63 0.21) (1.93 0.24; 0.012). No differences LC3B/A. size, (10,092.88 104.67μm2) larger CSA (7284.69 70.91μm2, 0.001) (5615.59 59.97μm2; 0.001), 70.91μm2) 0.001). CONCLUSION: supplementation could prevent fragmentation (reduction DRP1), thus, potentially resulting decreased demand fusion MFN2). In addition, greater rate autophagosome degradation formation LC3B) (indicative an increase macro-autophagy). Improvement contribute attenuating reduction size supplementation.

Dietary supplementation with shiikuwasha extract attenuates dexamethasone-induced skeletal muscle atrophy in aged rats

BACKGROUND Skeletal muscle atrophy is caused by a variety of diseases and conditions. In particular, skeletal muscle atrophy in the elderly contributes to a loss of independence with advanced age and increases the risk of falling. However, the effect of food consumed on a daily basis on skeletal muscle atrophy has been the focus of little research. In this study, the effects of dietary suppleme...

Mitochondrial signaling contributes to disuse muscle atrophy.

It is well established that long durations of bed rest, limb immobilization, or reduced activity in respiratory muscles during mechanical ventilation results in skeletal muscle atrophy in humans and other animals. The idea that mitochondrial damage/dysfunction contributes to disuse muscle atrophy originated over 40 years ago. These early studies were largely descriptive and did not provide uneq...

Isorhamnetin mitigates learning and memory disturbances in streptozotocin-induced diabetic rats

Background and Objective: Diabetes as a metabolic disorder can cause memory and learning impairment. In recent years, the effect of plant extracts on the treatment of diabetes mellitus has been raised. The purpose of this study was to determine the effect of isorhmnetin administration on learning and memory disability in an experimental model of streptozotocin-induced diabetes mellitus in rats....

EFFECT OF 6 WEEKS ENDURANCE TRAINING WITH QUERCETIN SUPPLEMENTATION ON THE EXPRESSION OF MITOCHONDRIAL PGC-1Α AND ATF2 GENES IN THE HIPPOCAMPUS OF DIABETIC MALE RATS

Background & Aims: Hyperglycemia causes disorders of cardiovascular system, kidneys, retina, lens of the eyes, skin, and central and peripheral nervous system. Since two indicators PGC-1α and Atf2 are involved in mitochondrial biogenesis, therefore, this study was designed and implemented to evaluate the effect of 6 weeks’ endurance training with quercetin supplementation on the expression of m...

Prophylactic effects of swimming exercise on autophagy-induced muscle atrophy in diabetic rats

Diabetes decreases skeletal muscle mass and induces atrophy. However, the mechanisms by which hyperglycemia and insulin deficiency modify muscle mass are not well defined. In this study, we evaluated the effects of swimming exercise on muscle mass and intracellular protein degradation in diabetic rats, and proposed that autophagy inhibition induced by swimming exercise serves as a hypercataboli...
